1. ⚙️ Metabolism and What Enzymes Areمیٹابولزم اور انزائمز کیا ہیں؟
🏷 Tags: Metabolism, Anabolism, catabolism, Enzymes, speed up
ENGLISH

Metabolism is all the chemical reactions in a cell/body. Anabolism builds big molecules from small ones (needs energy); catabolism breaks big molecules into small ones (releases energy). Both run all the time.

Enzymes are biological catalysts — usually proteins — that speed up chemical reactions without being used up themselves. Without enzymes, most body reactions would be far too slow for life. Example: amylase digests starch; pepsin digests proteins.

2. 🔑 Mechanism of Enzyme Action — Lock and Key
🏷 Tags: active site, substrate, enzyme–substrate complex, products, induced fit model, cofactors
ENGLISH

An enzyme has an active site — a shape that fits only its substrate (the molecule it acts on), like a lock fits its key. Steps: (1) substrate fits into the active site → enzyme–substrate complex; (2) the reaction happens (substrate changed); (3) products leave; (4) the enzyme is free again, unchanged, ready for the next substrate.

Because the fit is specific, each enzyme works on only one (or few) substrates — e.g. amylase only on starch. This specificity is why the lock-and-key model is used. The induced fit model adds that the active site reshapes slightly when the substrate binds, gripping it better.

Some enzymes also need cofactors — non-protein helpers — to work fully. Inorganic cofactors are metal ions (e.g. zinc, magnesium). Organic cofactors are of two kinds: prosthetic groups bind tightly to the enzyme (e.g. the haem group, some vitamins), while coenzymes bind loosely (e.g. many vitamins, NAD and NADP). Enzymes working inside cells are intracellular (e.g. respiratory enzymes); those secreted outside are extracellular (e.g. digestive enzymes in the stomach cavity).

3. 🌡️ Factors Affecting Enzyme Activity
🏷 Tags: Temperature, optimum, denatures, pH, Substrate concentration, Enzyme concentration
ENGLISH

Temperature: activity rises with temperature up to an optimum (about 37 °C in humans), then falls fast because the enzyme denatures (its shape is destroyed) at high temperature. pH: each enzyme has an optimum pH (pepsin ~pH 2 in the stomach; trypsin ~pH 8 in the intestine); wrong pH changes the active-site shape.

Substrate concentration: more substrate → faster reaction, until all active sites are full (saturation), then no further rise. Enzyme concentration: more enzyme → faster reaction (more active sites available).

4. 🚫 Enzyme Inhibition
🏷 Tags: Inhibitors, Competitive inhibitors, Non-competitive inhibitors
ENGLISH

Inhibitors are molecules that slow or stop enzymes. Competitive inhibitors look like the substrate and block the active site (they compete for it) — adding more substrate can overcome them. Non-competitive inhibitors bind elsewhere on the enzyme and change its shape, so the substrate no longer fits — adding more substrate does not help.

Inhibition is useful: many medicines work by inhibiting enzymes (e.g. some antibiotics and drugs), and cells use inhibition to control their own pathways (feedback inhibition).

Section B: Short Questions
1. Define metabolism. Differentiate between catabolism and anabolism. Which type of metabolism demands input of energy? Give an example.

Answer: Metabolism is all the biochemical reactions in living organisms. Anabolism builds large molecules from small ones (needs energy input, e.g. photosynthesis, protein synthesis); catabolism breaks large molecules into small ones (releases energy, e.g. respiration). Anabolism demands energy input.

2. Define an enzyme. What is its role in metabolism?

Answer: An enzyme is a biological catalyst, usually a protein, that speeds up a biochemical reaction without being used up. In metabolism it makes reactions fast enough for life and controls which reactions happen.

3. What is the active site of an enzyme? State its importance in enzyme specificity.

Answer: The active site is the region of the enzyme molecule where the substrate binds and catalysis happens. Its specific shape fits only its own substrate, which is why each enzyme is specific to one reaction.

4. Provide an example of an enzyme-substrate pair.

Answer: Amylase–starch (amylase breaks down starch), or pepsin–protein (pepsin breaks down proteins in the stomach).

5. What is the effect of substrate concentration on enzyme activity?

Answer: As substrate concentration rises, activity rises because more active sites are filled — until all sites are saturated, after which further substrate causes no more increase.

6. Provide two examples of enzymes that operate optimally at specific pH.

Answer: Pepsin works best at low pH (about pH 2, in the stomach); trypsin works best at high pH (about pH 8, in the small intestine).

7. What do you understand by optimum temperature and pH?

Answer: Optimum temperature and pH are the values at which an enzyme works at its maximum rate. Below or above them activity falls; extremes denature the enzyme.

8. What is the effect of enzyme concentration on enzyme activity?

Answer: More enzyme means more active sites available, so the reaction rate increases (as long as substrate is available).

9. Which enzyme inhibitors inhibit the enzymes without attaching to the active site?

Answer: Non-competitive inhibitors — they bind to a site other than the active site and change the enzyme's shape, so the substrate no longer fits.

10. Differentiate between competitive and non-competitive inhibition.

Answer: Competitive inhibitors resemble the substrate and block the active site (competition; more substrate can overcome them). Non-competitive inhibitors bind elsewhere, change the enzyme's shape, and cannot be overcome by adding more substrate.

Section C: Long Questions
1. Describe the characteristics of enzymes.

Answer: Enzymes are usually proteins; they are biological catalysts that speed up reactions without being used up; they are highly specific (one enzyme, one substrate/reaction); they work best at an optimum temperature and pH; they can be inhibited; some need cofactors; they work inside cells (intracellular) or outside (extracellular).

2. Describe how temperature extremes can inhibit enzyme activity and lead to enzyme denaturation.

Answer: At low temperature molecules move slowly, so enzyme–substrate collisions are rare and activity is low (but the enzyme is not destroyed). At high temperature activity first rises, then the enzyme's shape breaks down — it denatures — the active site is lost and activity falls sharply and permanently.

3. How does pH affect enzyme activity?

Answer: Each enzyme has an optimum pH at which its active site has the right shape and charge for its substrate. Changing the pH changes the charges on the enzyme, distorting the active site, so binding and catalysis fall. Extreme pH denatures the enzyme. Example: pepsin at pH 2, trypsin at pH 8.

4. Describe the factors that affect the activity of enzymes.

Answer: Temperature (rises to optimum, then denaturation), pH (optimum pH per enzyme), substrate concentration (rises to saturation), enzyme concentration (more enzyme, faster rate), inhibitors (competitive and non-competitive), and cofactors (needed by some enzymes for full activity).

5. Compare the Lock-and-Key and Induced Fit models of enzyme action.

Answer: Lock-and-Key: the active site has a fixed shape that exactly fits the substrate, like a lock fits its key; the enzyme does not change shape. Induced Fit: the active site is flexible — when the substrate binds, the active site reshapes slightly to grip it better, then catalysis happens. Both explain specificity; induced fit explains why binding improves fit.
