INTRODUCTION

Biochemistry is the chemistry of life: carbohydrates for energy, proteins for structure and catalysis, lipids for membranes and storage, nucleic acids for information, and minerals for health. This unit covers their structures, the four levels of protein structure, enzyme action and inhibition, and DNA's double helix.

1. Carbohydrates: Sugars and Polysaccharides
🏷 Tags: carbohydrate, glucose, starch, cellulose, glycogen, polysaccharide
ENGLISH

Carbohydrates are built from sugars. Glucose is the building block of cellulose, starch and glycogen — but NOT of inulin (inulin is a polymer of fructose). Aldoses have an aldehyde group (glucose, galactose, mannose); ketoses have a ketone group — so fructose is not an aldose.

Polysaccharides: starch (amylose + amylopectin, α-glucose, energy store in plants), glycogen (highly branched α-glucose, energy store in animals and humans — glycogen's role in the human body is to store glucose in the liver and muscles and release it when blood glucose falls), and cellulose (β-glucose, straight chains with hydrogen bonding, structural fibre in plant walls). Glucose vs fructose: glucose is an aldose (aldehyde at C-1), fructose is a ketose (ketone at C-2). Starch vs cellulose: same monomer (glucose) but different linkage (α-1,4/α-1,6 vs β-1,4) and shape (helical/branched vs straight fibres).

Carbohydrate factsGlucose builds: cellulose, starch, glycogen (NOT inulin)Aldoses: glucose, galactose, mannose; ketose: fructoseGlycogen: human glucose store (liver + muscles)Cellulose: β-glucose, straight H-bonded fibres
2. Proteins: Four Levels of Structure
🏷 Tags: protein, primary, secondary, tertiary, quaternary, denaturation
ENGLISH

Proteins are polymers of amino acids joined by peptide bonds. Primary structure: the amino-acid sequence. Secondary structure: α-helix and β-pleated sheet, held by hydrogen bonds between –CO and –NH groups. Tertiary structure: the 3D folding of one chain, held by hydrogen bonds, ionic bonds, disulfide bonds and hydrophobic interactions between R groups — tertiary structure is formed by folding driven by R-group interactions, and it creates the precisely shaped active site of an enzyme. Quaternary structure: several chains together (e.g. haemoglobin, 4 chains).

Denaturation breaks the bonds that hold the secondary and tertiary structure — hydrogen bonds (and other non-covalent interactions) — but NOT the peptide bonds, so the sequence stays intact while the shape (and function) is lost. All proteins contain C, H, N and O; sulphur is NOT present in all proteins (only those with cysteine/methionine).

Protein structure levelsPrimary: amino-acid sequenceSecondary: α-helix, β-sheet (H-bonds)Tertiary: 3D folding (R-group interactions) → active siteDenaturation breaks H-bonds, NOT peptide bonds
3. Enzymes: Action and Inhibition
🏷 Tags: enzyme, active site, lock and key, inhibitor, specificity
ENGLISH

Enzymes are biological catalysts (proteins) that speed up reactions without being used up. Mechanism: the substrate binds to the enzyme's active site (lock-and-key model, refined by induced fit), the enzyme–substrate complex forms, bonds are strained and broken/formed, products are released and the enzyme is unchanged.

Why is an enzyme specific? Only a substrate with the right shape and chemistry fits the active site — that is why an enzyme catalyses only a specific reaction. Competitive inhibition: an inhibitor resembling the substrate competes for the active site (overcome by more substrate). Non-competitive inhibition: the inhibitor binds elsewhere and changes the active-site shape permanently (not overcome by more substrate) — the book describes it as permanently changing the active site's shape, deactivating the enzyme.

Pepsin has an optimum pH of 2 because it works in the stomach, where the acid environment keeps its active site in the right shape; at higher pH it denatures. Enzyme inhibitors in medicine: many drugs are inhibitors — e.g. penicillin inhibits a bacterial enzyme, and ACE inhibitors block an enzyme that raises blood pressure.

Enzyme inhibitionCompetitive: competes for the active site (reversible)Non-competitive: binds elsewhere, changes active-site shapeSpecificity: only the right substrate fits the active sitePepsin optimum pH = 2 (stomach acid)
4. Lipids: Fats, Oils, Steroids and Membranes
🏷 Tags: lipid, fat, oil, saturated, unsaturated, steroid, membrane
ENGLISH

Fats and oils are triacylglycerols (triglycerides): glycerol + three fatty acids, joined by ester links. Fats are solid (saturated fatty acids, straight chains, pack tightly); oils are liquid (unsaturated fatty acids with cis double bonds that kink the chains and prevent packing).

Lipid classes: triacylglycerols (energy storage), phospholipids (membranes), glycolipids (carbohydrate + lipid, cell recognition) vs lipoproteins (lipid + protein, transport in blood), steroids (four fused rings), and waxes — esters of long-chain fatty acids and long-chain alcohols (not glycerol). Cell membranes are made of a phospholipid bilayer.

Steroids have four fused rings (three 6-membered, one 5-membered). Cholesterol is a steroid (a lipid, not a protein) found in membranes, where it controls fluidity; it is also the starting material for steroid hormones and vitamin D. Cholesterol is included in the lipid family because it is insoluble in water and soluble in non-polar solvents, like other lipids.

Lipid classesTriacylglycerols: glycerol + 3 fatty acids (ester links)Phospholipids: bilayer membranesWaxes: long-chain fatty acid + long-chain alcohol estersSteroids: four fused rings (cholesterol)
5. Nucleic Acids, DNA and Mineralsنیوکلک ایسڈز، DNA اور منرلز
🏷 Tags: nucleic acid, DNA, RNA, base pairing, minerals, iron
ENGLISH

DNA is a double helix: two strands joined by hydrogen bonds between nitrogenous bases — adenine pairs with thymine (2 hydrogen bonds), guanine with cytosine (3 hydrogen bonds). The backbone is made of phosphodiester bridges joining sugars. DNA stores genetic information; it is transcribed into mRNA, which is translated at ribosomes into proteins, with tRNA delivering amino acids.

DNA vs RNA: DNA is double-stranded with thymine; RNA is single-stranded with uracil instead of thymine. RNAs: mRNA (carries the code), tRNA (delivers amino acids), rRNA (ribosome structure). Complementary sequence: for T-A-C-C-G-A on one strand, the opposite strand reads A-T-G-G-C-T.

Minerals: potassium (nerve impulses, bananas, potatoes), iron (haemoglobin, red meat, spinach — iron in the body carries oxygen in haemoglobin), calcium (bones, teeth, dairy), magnesium (chlorophyll, enzymes, nuts), zinc (enzymes, immunity, meat, seeds). Fluorine (fluoride) prevents tooth decay by strengthening tooth enamel.

DNA base pairingA–T: 2 hydrogen bondsG–C: 3 hydrogen bondsBackbone: phosphodiester bridgesT-A-C-C-G-A pairs with A-T-G-G-C-T
DNA samjho: double helix base pairing سے جڑی رہتی ہے — A صرف T سے, G صرف C سے.

Q4. How the tertiary structure of proteins is formed?

Answer: The polypeptide chain folds into its 3D shape, driven by interactions between R groups: hydrogen bonds, ionic bonds, disulfide bridges and hydrophobic interactions. This folding creates the protein's specific shape, including enzyme active sites.

Q5. In what way fats and oils are different?

Answer: Both are triacylglycerols, but fats are solid at room temperature (saturated fatty acids, straight chains, pack well) while oils are liquid (unsaturated fatty acids with cis double bonds that kink the chains and prevent close packing).

Q6. Describe the structure of steroids. Write a note on the functions of cholesterol.

In medicine, many drugs are inhibitors: penicillin inhibits a bacterial wall enzyme; ACE inhibitors lower blood pressure; HIV protease inhibitors block viral replication.

Section 3: Constructed Response Questions
Q1. Why does the digestive enzyme pepsin have an optimum pH of 2?

Answer: Pepsin works in the stomach, where gastric juice has pH ≈ 2. Its active site is shaped correctly only in strong acid; at higher pH the enzyme denatures and loses activity. The optimum matches its working environment.

Q2. How condensation of sugars and condensation of amino acid are same?

Answer: Both are condensation reactions: two monomers join with the loss of a water molecule. Sugars join by glycosidic bonds (e.g. glucose + glucose → maltose + H₂O); amino acids join by peptide bonds (amino acid + amino acid → dipeptide + H₂O).

Q3. Why is cholesterol included in the lipid family?

Answer: Although it is not a fat, cholesterol is insoluble in water and soluble in non-polar solvents — the defining property of lipids — and it is built from the same isoprene-derived framework as other lipids. It also sits in membranes like other lipids.

Q4. Why fats and oils are also called triacylglycerols?

Answer: Because each molecule is glycerol (a triol) esterified with three fatty acids — three acyl groups attached to the glycerol backbone by ester links.

Q5. How hydrogen bonding is important in the double helix of DNA?

Answer: Hydrogen bonds between complementary bases (A–T with 2 bonds, G–C with 3) hold the two strands together, keep the helix stable, and allow the strands to separate for replication and transcription. The specific pairing also stores the genetic code.

Q6. If the sequence T-A-C-C-G-A located on one strand of DNA, what sequence would be seen opposite to it on the other strand?

Answer: A-T-G-G-C-T (A pairs with T, T with A, C with G, C with G, G with C, A with T). جواب: A-T-G-G-C-T۔

Section 4: Descriptive (Long) Questions
Q1. Classify and describe the structure of polysaccharides.

Answer: Homopolysaccharides of glucose: starch (plants: amylose, unbranched α-1,4; amylopectin, α-1,4 with α-1,6 branches — energy store), glycogen (animals: like amylopectin but more highly branched — rapid glucose release), cellulose (plants: β-1,4 straight chains, hydrogen-bonded fibres — structural). Also: chitin (N-acetylglucosamine, arthropod shells). Structure decides function: branching for storage, straight fibres for strength.

Q2. How the structure of proteins support its function. Describe at each structure level.

Answer: Primary: the sequence decides all higher levels (e.g. sickle-cell anaemia from one amino-acid change). Secondary: helices/sheets give local stability. Tertiary: folding creates the active site (enzymes), binding pockets (antibodies) or fibres (collagen). Quaternary: multiple chains allow cooperation (haemoglobin's 4 chains bind oxygen cooperatively).

Q3. Write an account of classification of lipids with suitable examples.

Answer: Simple lipids: fats and oils (triacylglycerols), waxes (long-chain esters). Compound lipids: phospholipids (membranes), glycolipids (cell recognition). Derived lipids: steroids (cholesterol, hormones), fat-soluble vitamins (A, D, E, K). Functions: energy storage, membranes, insulation, signalling, vitamin carriers.

Q4. What are nucleic acids? Describe the role of DNA and RNA in the protein synthesis.

Answer: Nucleic acids are polymers of nucleotides (sugar + phosphate + base). DNA stores the genetic code in its base sequence in the nucleus. Transcription copies one strand into mRNA; mRNA travels to ribosomes; translation reads codons (3 bases = 1 amino acid), tRNA delivers the amino acids, rRNA forms the ribosome — producing the protein specified by the DNA.
