Biotechnology is a branch of biology that deals with the manipulation of organisms at molecular and cellular level to get services and products beneficial for human welfare. It benefits human health, agriculture, industries and the environment. Genetic engineering (genetic modification) is its core tool: changing an organism's DNA deliberately to add a useful trait.
Bacteria are commonly used in genetic engineering because they grow quickly and accept human genes — a gene inserted into bacteria can be copied millions of times as the bacteria multiply.
Steps to transfer a human gene into a bacterial cell: (1) Cutting the desired gene from DNA — the first step — using special enzymes. (2) Insert the gene into a vector — usually a plasmid (a small circular DNA of bacteria) or a virus. (3) Introduce the recombinant plasmid into a bacterium. (4) After inserting the gene, the host cell is allowed to multiply and express the gene. The joined DNA from two sources is called recombinant DNA.
Plasmids are used because they are small, circular, easy to cut and join, and replicate inside bacteria — ideal gene carriers. The book's diagram labels, in order: 1 = desired gene cut from donor DNA, 2 = gene fragment, 3 = plasmid from the bacterium, 4 = recombinant plasmid (gene + plasmid), 5 = transformed bacterium producing the product.
Food/agriculture: crops improved by genetic engineering — pest resistance (a gene for toxin production makes plants resist insect attacks), higher yield, better nutrition. Golden rice is genetically modified to increase vitamin A. Medicine: genetically modified bacteria produce insulin (diabetes), growth hormone and interferon; vaccines (not antibiotics) are the biotechnology products that help against viral infections — biotechnology also helped develop vaccines quickly, like for COVID-19. Gene editing can correct faulty genes and improve crops and treatments precisely.
Yeast in bread: fermentation produces carbon dioxide that raises dough; in ethanol production, yeast ferments sugars from crops (corn, sugarcane) into ethanol, which can be blended with petrol as a cleaner fuel. Fermented plant starch gives biodegradable plastic (bioplastic); microbial fermentation gives antibiotics such as penicillin (from Penicillium mould) and human insulin through bacteria.
Health concerns: some people worry genetically modified foods could cause allergic reactions or long-term health problems, and that new genes might interact with human cells in unexpected ways — which is why GM foods' effects on health must be studied carefully. Environmental impact: GM plants can cross-pollinate with wild plants, spreading modified genes into natural ecosystems. Loss of biodiversity: if farmers mostly grow GM crops, traditional varieties may become rare or extinct. Resistant pests and weeds: pests repeatedly exposed to GM crops may evolve into "powerful pests" no longer affected by the crop's built-in defences. Ethical issues: many people feel it is morally wrong to alter the natural genes of living organisms.

1. What is the relation between biotechnology and genetic engineering?
Answer: Biotechnology is the broad field of using organisms (or their parts) at molecular and cellular level for useful products and services. Genetic engineering is a tool within biotechnology — deliberately changing an organism's DNA — used to create GM organisms and products.
2. What is a plasmid? Why biologists use plasmids in genetic engineering?
Answer: A plasmid is a small, circular, extra DNA molecule found in bacteria, separate from the main chromosome. Biologists use plasmids as vectors: they are easy to cut open, join with a desired gene, put back into bacteria, and copy as the bacteria multiply — so they carry foreign genes into host cells.
3. The following diagram shows how a gene is transferred to bacterial cell. Identify the structures labelled as 1 to 5.
Answer: 1 = desired gene cut from donor-cell DNA. 2 = the cut gene fragment. 3 = plasmid taken from the bacterium. 4 = recombinant plasmid (plasmid + desired gene joined). 5 = transformed bacterium that multiplies and produces the gene product.
4. What do you mean by genetically modified organism?
Answer: A genetically modified organism (GMO) is an organism whose DNA has been deliberately changed by genetic engineering — e.g. a gene from another organism inserted — so it gains a new useful trait such as pest resistance or extra vitamin production.
5. List two types of medical products that can be produced using genetic engineering.
Answer: Human insulin (for diabetes) and human growth hormone. (Also: interferon, clotting factors and vaccines such as hepatitis B vaccine.)
6. List two ways by which genetic engineering may improve crops.
Answer: (1) Pest resistance — e.g. a toxin gene makes plants resist insect attacks, reducing pesticide use. (2) Better nutrition or yield — e.g. golden rice with more vitamin A, or crops that tolerate drought/disease and give higher harvests.
1. Explain with examples that food biotechnology has advanced agriculture.
Food biotechnology has advanced agriculture by GM crops: Bt crops carry a toxin gene so insects eating them die, cutting pesticide use and raising yield; golden rice carries genes for vitamin A production, fighting deficiency; crops are engineered for disease resistance, drought tolerance and longer shelf life, so farmers harvest more, more reliably, with fewer chemicals. Fermentation biotechnology also gives bread, yogurt, cheese and ethanol from crops.
2. Explain with examples that medical biotechnology has advanced healthcare in diabetes and cancer.
Diabetes: human insulin is now made by GM bacteria carrying the human insulin gene — pure, plentiful and safer than animal insulin; patients control blood sugar reliably. Cancer: biotechnology gives targeted therapies and interferon, diagnostic gene tests that detect cancer-related genes early, and vaccines (e.g. HPV vaccine) that prevent virus-caused cancers; gene research is developing therapies that attack cancer cells specifically.
3. Explain the role of yeast in the production of bread and ethanol.
Yeast is a fungus used in fermentation. In bread-making, yeast ferments sugars in dough, producing carbon dioxide gas that forms bubbles and raises the dough, plus flavours. In ethanol production, yeast ferments sugars from crops (corn, sugarcane) into ethanol and carbon dioxide; the ethanol is distilled and can be blended with petrol as a cleaner-burning fuel.
4. Enlist the steps for transferring a human gene into a bacterial cell.
(1) Identify and cut the desired human gene from donor DNA with enzymes. (2) Cut open a bacterial plasmid (vector) with the same enzymes. (3) Join the gene into the plasmid to form recombinant DNA. (4) Introduce the recombinant plasmid into a bacterium (transformation). (5) Grow the transformed bacteria; they multiply and express the gene, producing the human protein (e.g. insulin), which is then collected and purified.
5. Describe the advantages of gene editing.
Gene editing can precisely correct faulty genes that cause inherited diseases; create crops that resist pests, disease and drought with less pesticide; increase food nutrition (e.g. vitamin A rice); produce medicines like insulin cheaply in microbes; develop faster vaccines; and help endangered species or remove disease-carrying traits. Precision means fewer unwanted changes than older breeding methods.
6. Discuss the potential risks of genetic modification.
Health: possible allergies or unknown long-term effects of GM foods; new genes might interact with human cells unexpectedly. Environment: GM genes can spread to wild relatives by cross-pollination and disturb ecosystems. Biodiversity: dominance of a few GM varieties can make traditional varieties extinct. Pests/weeds: repeated exposure can evolve resistant "powerful pests". Ethics and control: moral objections to altering natural genes, and questions about who controls GM seeds and technology.
1. How can inserting a gene into a plant make it pest-resistant?
Answer: The inserted gene (e.g. from the bacterium Bacillus thuringiensis, Bt) makes the plant produce a toxin protein in its tissues. When pest insects eat the plant, the toxin damages their gut and kills them, while the plant itself is unharmed — so the plant defends itself without spraying.
2. How can biotechnology help in developing vaccines quickly, like for COVID-19?
Answer: Once a virus's gene sequence is known, biotechnology platforms (e.g. mRNA) can be programmed to make a harmless piece of the virus (like its spike protein) without growing the live virus. Production can start immediately, scale up in bioreactors, and be updated fast if the virus changes — much quicker than traditional vaccine methods.
3. Why is it important to carefully study the effects of genetically modified foods on health?
Answer: Because a new gene means a new protein in food, which could in principle trigger allergy or interact with human biology in unforeseen ways, and effects might appear only after long-term eating. Careful testing — for allergenicity, toxicity and nutrition — before approval, plus monitoring afterwards, protects consumers and builds trust.

پھر applications کی تین groups بناؤ: food (golden rice vitamin A), medicine (insulin, growth hormone, interferon, vaccines), industry (yeast fermentation).
