INTRODUCTION

Synthesis is planning: how do we build a target molecule from simple starting materials? This unit introduces retrosynthesis — working backwards with disconnections, synthons and synthetic equivalents — functional group interconversion (FGI), and applies the ideas to real drugs: paracetamol, aspirin, and the AI-discovered antibiotic Halicin.

1. Synthesis vs Retrosynthesis
🏷 Tags: synthesis, retrosynthesis, target molecule, disconnection, synthon
ENGLISH

Synthesis builds a target molecule forward, step by step, from starting materials. Retrosynthesis works backwards: start from the target molecule and break it apart (a disconnection, shown by the ⇒ arrow) into simpler pieces, until you reach cheap, available starting materials. Then you reverse the arrows to get the real synthesis.

After disconnection, the pieces obtained are called synthons — idealised fragments, often charged (e.g. ⁺CH₃ or ⁻OH), which may not exist on their own. A synthetic equivalent is the real reagent that behaves like the synthon (e.g. CH₃Cl is the synthetic equivalent of ⁺CH₃; NaOH of ⁻OH).

🏷 Tags: FGI, functional group interconversion, reagent, conversion
ENGLISH

The primary purpose of Functional Group Interconversion (FGI) is to change a functional group into another to enable a specific reaction. A group that will not react the way you need is converted into one that will. FGI does not break the molecule into smaller fragments and does not add carbon atoms — it just swaps the functional group.

Key conversions (the toolkit): alcohol → halogenoalkane (SOCl₂, PCl₅, HBr); halogenoalkane → amine (NH₃), alkene (ethanolic KOH), nitrile (KCN); alkene → alcohol (steam/H₃PO₄); alcohol → aldehyde/ketone/acid (oxidation, K₂Cr₂O₇); aldehyde/ketone → alcohol (NaBH₄); carboxylic acid → alcohol (LiAlH₄); nitrile → amine (reduction, LiAlH₄); nitro group → amino group (Sn/HCl).

One useful application: converting a carboxylic acid group into an amide group (via the acyl chloride) so it can be reduced to an amine — or converting an alcohol into a halogenoalkane so it can be substituted by an amine, an alkene or a nitrile.

FGI toolkitAlcohol → halogenoalkane (SOCl2, HBr)Halogenoalkane → amine (NH3), alkene (KOH), nitrile (KCN)Aldehyde/ketone → alcohol (NaBH4)Nitrile → amine (LiAlH4); nitro → amino (Sn/HCl)
3. Paracetamol and Aspirin Retrosynthesis
🏷 Tags: paracetamol, aspirin, retrosynthetic tree, starting material
ENGLISH

Paracetamol (4-hydroxyacetanilide): retrosynthesis disconnects the amide bond → 4-aminophenol + ethanoic anhydride (synthetic equivalents). Forward synthesis: phenol → (nitration) 4-nitrophenol → (reduction, Sn/HCl) 4-aminophenol → (acetylation with ethanoic anhydride) paracetamol. The exercise sequence shows exactly this: phenol → nitrophenol → aminophenol → acetylation → paracetamol.

Aspirin (acetylsalicylic acid): retrosynthesis disconnects the ester bond → salicylic acid + acetic anhydride (the two synthetic equivalents). Forward: salicylic acid + acetic anhydride (acid catalyst) → aspirin + ethanoic acid. Retrosynthetic trees for both drugs start from the drug and branch back to phenol/benzene-level starting materials.

4. Halogenoalkanes in Retrosynthesis
🏷 Tags: halogenoalkane, synthon, synthetic equivalent, route
ENGLISH

Halogenoalkanes are the key intermediates in retrosynthesis because the C–X bond is a versatile handle: from a halogenoalkane you can make amines (NH₃), alkenes (ethanolic KOH), nitriles (KCN, adding a carbon), alcohols (aqueous NaOH) and more. So when planning a synthesis, a common move is to disconnect back to a halogenoalkane, then back to the alcohol or alkane it came from.

Worked conversions: CH₃CH₃ → CH₃COOH: ethane → (Cl₂/UV) chloroethane → (NaOH aq) ethanol → (oxidation) ethanal → (oxidation) ethanoic acid. CH₂=CH₂ → CH₃CH₂OH: hydration with steam/H₃PO₄. CH₃CH₂CH₂Cl → CH₃CH=CH₂: hot ethanolic KOH (elimination). CH₃COOH → CH₃CH₂OH: LiAlH₄ reduction.

5. AI in Drug Discovery: Halicinڈرگ ڈسکوری میں AI: ہیلیسن
🏷 Tags: Halicin, AI, drug discovery, screening, antibiotic
ENGLISH

Halicin is an antibiotic discovered by an MIT machine-learning model that screened thousands of candidate molecules for antibacterial activity. It works as an antibiotic drug — effective even against drug-resistant bacteria. Halicin shows how AI can find drugs that human chemists might miss, by predicting activity from molecular structure rather than testing one compound at a time.

Safety note (book-faithful): this unit describes Halicin as a discovery example only. Making or testing antibiotics is laboratory work for trained professionals — the guide-book point is the retrosynthesis and AI-screening idea, not a home procedure.

Section 4: Descriptive (Long) Questions
Q1. Give a detailed account of importance of retrosynthesis in organic synthesis.

Answer: Retrosynthesis turns an unstructured problem ("how do I make this?") into a search: by disconnecting bonds that correspond to known reactions, chemists find short, cheap, high-yield routes, avoid dead ends, and choose starting materials that are available. It is essential for complex targets like drugs, where many routes are possible but only a few are practical, safe and economical.

Q2. Explain the role of halogenoalkanes in retrosynthesis of organic molecules.

Answer: Halogenoalkanes are versatile branch points: their C–X bond can be replaced by –NH₂, –CN, –OH, or eliminated to an alkene, and the carbon skeleton can be extended (via cyanide). So syntheses are routinely disconnected back to a halogenoalkane, which is itself made from an alcohol or alkene — making the halogenoalkane the hub of most synthetic plans.

Q3. Construct the retrosynthetic tree for the aspirin and design its synthesis.

Answer: Retrosynthesis: aspirin ⇒ (break ester bond) salicylic acid + acetic anhydride; salicylic acid ⇒ phenol (Kolbe reaction). Forward: phenol → (NaOH, CO₂, Kolbe) salicylic acid → (acetic anhydride, acid catalyst) aspirin + ethanoic acid.

Q4. Design the synthesis of the following molecules starting from benzene. a) phenylamine b) toluene c) benzoic acid d) benzyl alcohol

Answer: a) Phenylamine: benzene → (HNO₃/H₂SO₄) nitrobenzene → (Sn/HCl) phenylamine. b) Toluene: benzene → (CH₃Cl/AlCl₃) toluene. c) Benzoic acid: benzene → (CH₃Cl/AlCl₃) toluene → (KMnO₄/H⁺) benzoic acid. d) Benzyl alcohol: benzene → toluene → (Cl₂, UV) benzyl chloride → (NaOH aq) benzyl alcohol.

Q5. How will you carry out the following conversions? a) CH₃–CH₃ → CH₃–COOH b) CH₂=CH₂ → CH₃–CH₂–OH c) CH₃–CH₂–CH₂–Cl → CH₃–CH=CH₂ d) CH₃–COOH → CH₃–CH₂–OH

Answer: a) Ethane → (Cl₂/UV) chloroethane → (NaOH aq) ethanol → (K₂Cr₂O₇, reflux) ethanoic acid. b) Ethene + steam/H₃PO₄ (hydration) → ethanol. c) 1-chloropropane + hot ethanolic KOH → propene (elimination). d) Ethanoic acid + LiAlH₄ (dry ether), then acid work-up → ethanol.
