INTRODUCTION

Nitrogen brings some of the most useful chemistry in the book: amines (bases and nucleophiles), diazonium salts (the gateway to dyes and phenols), nitriles, amides, and amino acids — the building blocks of proteins. This unit also explains why phenylamine is a weaker base than ammonia, and how penicillin works.

1. Amines: Classification and Basicity
🏷 Tags: amine, primary, secondary, tertiary, basicity, lone pair
ENGLISH

Amines are classified by how many carbon groups are attached to nitrogen: primary R–NH₂, secondary R₂NH (general formula RNHR), tertiary R₃N. Primary, secondary and tertiary amines with the same formula are functional group isomers of each other. Four isomeric amines of formula C₃H₉N are: propylamine (CH₃CH₂CH₂NH₂), isopropylamine ((CH₃)₂CHNH₂), ethylmethylamine (CH₃CH₂NHCH₃) and trimethylamine ((CH₃)₃N).

Basicity order (aqueous): secondary alkyl amines (e.g. diethylamine) > primary alkyl amines (e.g. ethylamine) > ammonia > phenylamine. Alkyl groups donate electrons (+I effect), making the nitrogen lone pair more available — so ethylamine is a stronger base than ammonia. Phenylamine is the weakest because its nitrogen lone pair delocalises into the benzene ring and is less available to accept a proton.

Amides (RCONH₂) are much weaker bases than ammonia because the nitrogen lone pair delocalises into the carbonyl group (resonance), so it is barely available for protonation.

Basicity order (aqueous)Diethylamine (2°) > ethylamine (1°) > ammoniaPhenylamine: weakest (lone pair delocalised into ring)Amides: much weaker than ammonia (pair in C=O)Alkyl groups donate electrons (+I) → stronger base
2. Preparation of Amines
🏷 Tags: preparation, reduction, nitro, nitrile, Gabriel
ENGLISH

Primary amines: (1) reduction of nitriles: R–CN + LiAlH₄ (or H₂/Ni) → R–CH₂NH₂ (e.g. ethanenitrile → ethylamine). (2) Reduction of amides: R–CONH₂ + LiAlH₄ → R–CH₂NH₂ (primary amine). (3) Reduction of nitro compounds: nitrobenzene + Sn/HCl → phenylamine. (4) Halogenoalkane + excess ammonia (ethanolic): R–X + NH₃ → R–NH₂.

Secondary amines: (1) halogenoalkane + a primary amine (careful: further alkylation can occur, so use excess amine); (2) reduction of N-substituted amides. Phenylamine preparations (two): nitrobenzene + Sn/HCl (or H₂/Ni), and the Hofmann degradation of benzamide (not in the book's core list — the standard school pair is nitro reduction and nitrile/amide reduction routes).

From ethanamine to methanamine (chain shortening): ethanamine → (HNO₂) ethanol → (oxidation) ethanoic acid → (NH₃, then heat) ethanamide → (Hofmann degradation, Br₂/NaOH) methanamine. This is the classic "descending the series" sequence.

3. Reactions of Amines and Diazotisation
🏷 Tags: reactions, nitrous acid, diazotisation, diazonium, azo dye
ENGLISH

With acids: amines form salts (R–NH₂ + HCl → R–NH₃⁺Cl⁻). With bromine water: phenylamine gives 2,4,6-tribromophenylamine (white precipitate) — like phenol, the ring is activated, this time by the –NH₂ lone pair. With nitrous acid (NaNO₂ + dilute HCl, below 10 °C): primary aromatic amines give diazonium salts; primary aliphatic amines give nitrogen gas; secondary amines give a yellow oily nitrosamine; tertiary amines just form a soluble salt with no visible change.

Diazonium chemistry: benzenediazonium chloride + phenol in NaOH → an azo compound (dye, –N=N– linkage). Benzenediazonium chloride heated with water → phenol + N₂ gas (the gas evolved is nitrogen). Benzenediazonium chloride + H₃PO₂ → benzene (N₂⁺ replaced by H) — used in the 1,3,5-tribromobenzene synthesis.

Nitrous acid test for aminesPrimary aromatic → diazonium salt (dye chemistry)Primary aliphatic → N2 gas evolvedSecondary → yellow oily nitrosamineTertiary → soluble salt, no visible change
4. Nitriles, Amides and Amino Acids
🏷 Tags: nitrile, amide, amino acid, zwitterion, peptide bond
ENGLISH

Nitriles: acid hydrolysis of R–CN gives a carboxylic acid (R–COOH + NH₃). Reduction of R–CN gives a primary amine (Unit: ethanenitrile → ethylamine). Amides: reduction of R–CONH₂ with LiAlH₄ gives a primary amine; hydrolysis gives the carboxylic acid and ammonia.

Amino acids are the monomers of proteins. In aqueous solution they exist as zwitterions — a dipolar form where –COOH has donated its proton to –NH₂, giving –NH₃⁺ and –COO⁻ in the same molecule. That is why an aqueous solution of glycine is neutral.

Peptide bond: the amide link (–CO–NH–) between two amino acids, formed by condensation (loss of water). A tripeptide (3 amino acids) has 2 peptide bonds. From glycine (Gly) and alanine (Ala) the possible tripeptides are: Gly-Gly-Gly, Gly-Gly-Ala, Gly-Ala-Gly, Ala-Gly-Gly, Gly-Ala-Ala, Ala-Gly-Ala, Ala-Ala-Gly, Ala-Ala-Ala (8 in total, 2³).

5. Penicillin, Azo Dyes and Electrophoresis
🏷 Tags: penicillin, beta-lactam, azo dye, electrophoresis
ENGLISH

Penicillin's key feature is its strained four-membered beta-lactam ring. The ring binds and inactivates transpeptidase, the enzyme that builds bacterial cell walls — so the bacterium cannot complete its wall and dies. Humans are unaffected because we have no cell walls. The chromophore in azo dyes is the –N=N– group: its extended conjugated system absorbs visible light, giving intense colour; changing the groups around it changes the colour.

Electrophoresis separates charged molecules (e.g. amino acids, proteins) by their movement in an electric field through a gel or buffer. The speed depends on charge and size: at a given pH, amino acids exist as cations, anions or zwitterions depending on their isoelectric point, so they move towards different electrodes at different speeds — used to identify and separate amino acids and proteins.

Section 2: Short Answer Questions
Q1. Give two preparations of phenyl amine.

Answer: (1) Nitrobenzene + Sn/concentrated HCl (then NaOH work-up) → phenylamine. (2) Nitrobenzene + H₂/Ni (catalytic hydrogenation) → phenylamine.

Answer: (1) A halogenoalkane + a primary amine (with excess amine to limit further alkylation). (2) Reduction of an N-substituted amide with LiAlH₄.

Q3. What is the chromophore in azo dyes? What is its function in dyes?

Answer: The chromophore is the –N=N– azo group (with its conjugated aromatic system). It absorbs visible light of a particular wavelength, giving the dye its colour; the rest of the molecule tunes the shade.

Q4. Why amides are less basic than ammonia.

Answer: In an amide the nitrogen lone pair is delocalised into the carbonyl group by resonance (R–C(=O)–NH₂ ↔ R–C(O⁻)=NH₂⁺), so it is much less available to accept a proton than ammonia's lone pair.

Q5. What is zwitter ion?

Answer: A dipolar ion: a molecule carrying both a positive and a negative charge but neutral overall, e.g. amino acids as ⁺H₃N–CHR–COO⁻.

Q6. Differentiate between primary, secondary & tertiary amines.

Answer: Primary: nitrogen bonded to 1 carbon group (R–NH₂). Secondary: 2 carbon groups (R₂NH). Tertiary: 3 carbon groups (R₃N). They differ in basicity (2° > 1° > 3° in some solvents, but aqueous order follows the book: secondary strongest), in reactions with nitrous acid, and in hydrogen bonding.

Q7. Write names & structures of four isomeric amines having the molecular formula C₃H₉N.

Answer: Propylamine CH₃CH₂CH₂NH₂; isopropylamine (CH₃)₂CHNH₂; ethylmethylamine CH₃CH₂NHCH₃; trimethylamine (CH₃)₃N.

Q8. Convert ethanamine into methanamine.

Answer: Ethanamine → (HNO₂) ethanol → (oxidation, K₂Cr₂O₇) ethanoic acid → (NH₃, heat) ethanamide → (Br₂/NaOH, Hofmann degradation) methanamine. Each step shortens the chain by one carbon at the amide stage.

Q9. How will you synthesize ethanal from ethanenitrile?

Answer: Ethanenitrile (CH₃CN) reduced with DIBAL-H or SnCl₂/HCl (Stephen reduction), then hydrolysed, gives ethanal (CH₃CHO). (Partial reduction of the nitrile stops at the aldehyde.)

Q10. Define peptide bond. How many peptide bonds are there in a tripeptide?

Answer: A peptide bond is the amide link (–CO–NH–) between the carboxyl group of one amino acid and the amino group of the next, formed by condensation. A tripeptide (3 amino acids) has 2 peptide bonds.

Q11. Give all the tripeptides that could possibly be synthesized from glycine (Gly), alanine (Ala).

Answer: 2³ = 8: Gly-Gly-Gly, Gly-Gly-Ala, Gly-Ala-Gly, Ala-Gly-Gly, Gly-Ala-Ala, Ala-Gly-Ala, Ala-Ala-Gly, Ala-Ala-Ala. جواب: کل 8: Gly-Gly-Gly، Gly-Gly-Ala، Gly-Ala-Gly، Ala-Gly-Gly، Gly-Ala-Ala، Ala-Gly-Ala، Ala-Ala-Gly، Ala-Ala-Ala۔

Section 3: Constructed Response Questions
Q1. Complete the following reaction sequence by giving the structures of the compounds. 1,3,5-tribromobenzene from benzene. Benzene → (HNO₃/H₂SO₄) → (A) → (Sn/HCl) → (B) → (Br₂ excess/FeBr₃) → (C) → (NaNO₂/HCl) → (D) → (H₃PO₂) → (E).

Answer: A = nitrobenzene. B = phenylamine (aniline). C = 2,4,6-tribromophenylamine (the –NH₂ group strongly activates the ring, so all three positions brominate). D = 2,4,6-tribromobenzenediazonium chloride. E = 1,3,5-tribromobenzene (H₃PO₂ replaces –N₂⁺ by –H).

Q2. 1,3,5-Tribromobenzene from benzene. Benzene → (HNO₃/H₂SO₄) → (A) → (Cl₂/FeCl₃) → (B) → (Sn/HCl) → (C) → (NaNO₂/HCl) → (D) → (H₂O) → (E).

Answer: NOTE: the book repeats the heading "1,3,5-Tribromobenzene" for this second scheme, but its steps actually give a different product: A = nitrobenzene; B = 3-chloronitrobenzene (–NO₂ is meta-directing, so Cl goes meta); C = 3-chloroaniline; D = 3-chlorobenzenediazonium chloride; E = 3-chlorophenol (diazonium + warm water → phenol). The scheme as printed leads to 3-chlorophenol, not 1,3,5-tribromobenzene.

Q3. Keep in view the scheme, devise a reaction sequence to prepare 4-Hydroxybenzoic acid from benzene.

Answer: Benzene → (Br₂/FeBr₃) bromobenzene → (NaOH, high temperature and pressure, Dow process) phenol → (KOH) potassium phenoxide → (CO₂, high temperature, Kolbe–Schmitt) potassium 4-hydroxybenzoate (para product favoured with K⁺ at higher temperature) → (dilute acid) 4-hydroxybenzoic acid.

Section 4: Descriptive (Long) Questions
Q1. Describe the relative basicity of aqueous ammonia, ethyl amine and phenyl amine.

Answer: Ethylamine > ammonia > phenylamine. Ethylamine: the ethyl group donates electrons (+I), making the lone pair more available than in ammonia. Phenylamine: the lone pair delocalises into the benzene ring, making it far less available. In water, the measured Kb order follows this explanation.

Q2. Give two preparation methods of each of the following. i) Primary amines ii) Secondary amines

Answer: i) Primary: reduction of nitriles (LiAlH₄) and reduction of nitro compounds (Sn/HCl); also halogenoalkane + excess ammonia. ii) Secondary: halogenoalkane + primary amine, and reduction of N-substituted amides (LiAlH₄).

Q3. Write a note on electrophoresis.

Answer: Electrophoresis separates charged species in an electric field. A sample (e.g. amino acids or proteins) is placed on a gel or paper in buffer and a voltage is applied: cations move to the cathode, anions to the anode, at speeds depending on charge and size. At a given pH an amino acid's charge depends on its isoelectric point, so different amino acids separate into bands that can be stained and identified — used in biochemistry and forensics.

Q4. Explain the formation of azo compounds from phenylamine and illustrate the preparation of a dye using this?

Answer: Phenylamine + NaNO₂ + dilute HCl below 10 °C → benzenediazonium chloride (diazotisation). The diazonium salt couples with phenol in alkaline solution (or with another aromatic amine) → an azo compound (C₆H₅–N=N–C₆H₄OH, orange dye). The –N=N– chromophore absorbs visible light. This is how azo dyes (e.g. methyl orange, p-hydroxyazobenzene) are made industrially.
