Genetic engineering is the artificial changing of an organism's DNA — cutting a gene out, and inserting it into another organism so that the new host makes the gene's product. An organism changed this way is a genetically modified organism (GMO): genes may have been introduced, enhanced or deleted.
Tools: Restriction endonucleases ('molecular scissors') cut DNA at specific recognition sequences, often leaving single-stranded sticky ends that match each other; they are also used to produce RFLPs (restriction fragment length polymorphisms — different fragment patterns used in DNA fingerprinting). DNA ligase ('molecular glue') joins DNA pieces — it does not cut DNA. Reverse transcriptase makes cDNA from mRNA (useful when the gene of interest is only available as mRNA). Gel electrophoresis separates DNA fragments by size: smaller fragments move further through the gel.
Vectors carry the gene into the host cell. The commonest vector is a bacterial plasmid — a small circular DNA that replicates inside bacteria and can carry a foreign gene with it. Bacterial cells are the ideal expression system for cloning because they grow fast, are cheap, and their genetics is well understood.
Step 1 — Obtain the gene of interest. It can be cut from a chromosome with restriction enzymes, made from mRNA by reverse transcription (cDNA), or built by artificial gene synthesis. Step 2 — Select and isolate it (e.g. by gel electrophoresis and probes that match its sequence).
Step 3 — Cut the vector with the same restriction enzyme, so its sticky ends match the gene's ends. Step 4 — Join them: mix gene and vector; sticky ends pair up, and DNA ligase seals them — the result is recombinant DNA (gene + vector). Step 5 — Introduce it into the host (transformation: bacteria take up the plasmid; also electroporation — electric shock opens pores — or gene gun for plant cells). Step 6 — Grow the host and harvest the product: host cells multiply (cloning the gene) and express it, making the desired protein, which is purified.
PCR vs recombinant DNA technology: PCR copies a DNA segment many times in a test tube (amplification, no host cell needed); recombinant DNA technology inserts a gene into a living host so the host copies and expresses it. Both are 'cloning' in a broad sense, but they work differently.
PCR is a test-tube technique that copies a chosen DNA segment millions of times. It needs: template DNA, two primers (short DNA pieces matching the segment's ends), DNA polymerase (heat-stable Taq polymerase, from a hot-spring bacterium), nucleotides, and a thermal cycler that changes temperature in cycles.
Each cycle has three steps: (1) Denaturation (~94–98 °C): the double helix separates into single strands. (2) Annealing (~50–60 °C): primers bind to their matching sequences. (3) Extension (72 °C, the optimum for Taq): Taq polymerase builds new strands from the primers. One cycle doubles the DNA; about 30 cycles make over a billion copies from a tiny starting sample.
Uses: Detecting infections (including viruses) from tiny samples, DNA fingerprinting in forensics and paternity testing, diagnosing genetic diseases, studying ancient DNA, and preparing genes for cloning. PCR is also the basis of many rapid diagnostic tests.
Transgenic bacteria: Bacteria carrying human genes make medicines cheaply and in bulk — insulin, human growth hormone, interferon and vaccine proteins. Transgenic animals: Animals with added genes grow faster, resist disease, or produce medicines in their milk ('pharming'). Transgenic plants: Crops with added genes resist pests (Bt cotton makes a bacterial toxin harmless to humans but deadly to bollworms), tolerate herbicides or drought, stay fresh longer, or carry extra nutrition (golden rice makes vitamin A precursor).
Role of GMOs in biotechnology products: GMOs are living factories — once the gene is in, the organism copies it with every division and makes the product continuously, which is far cheaper than extracting tiny amounts from human or animal tissue. That is why modern insulin, many vaccines and industrial enzymes come from GMOs.
Concerns: Gene flow to wild relatives, effects on non-target insects, allergy risks, and corporate control of seeds are debated; that is why GMOs are tested and regulated before release.
Before genetic engineering, insulin for diabetes came from pig and cow pancreases — expensive, in short supply, and sometimes causing allergic reactions. The modern method uses Escherichia coli:
The human insulin gene is obtained (from mRNA by reverse transcription, or synthesised). Because insulin has two chains (A and B), the genes for the two chains are inserted into separate plasmids (or as one joined gene) and introduced into E. coli by transformation. The bacteria grow in large fermenters and produce the chains, which are harvested, purified and joined by disulphide bonds into active human insulin — identical to the body's own insulin, available in large amounts at lower cost.
Introducing human DNA into bacteria — methods: (1) Transformation — bacteria made 'competent' (e.g. calcium chloride treatment) take up plasmid DNA. (2) Electroporation — a brief electric pulse opens temporary pores in the cell membrane. (3) Gene gun / microinjection — mainly for plant/animal cells, but the same idea: physically delivering DNA. Antibiotic-resistance genes on the plasmid let scientists select only the bacteria that received it.

Answer: (i) Endonuclease (restriction enzyme) cuts DNA at specific sites; ligase joins DNA fragments together. (ii) PCR amplifies DNA copies in a test tube without a host cell, using temperature cycles; recombinant DNA technology inserts a gene into a living host cell (via a vector) so the host replicates and expresses it.

Steps: (1) obtain the gene (cut from chromosome, cDNA from mRNA, or synthesis); (2) select and isolate it; (3) cut the vector with the same enzyme; (4) join gene and vector with ligase to make recombinant DNA; (5) introduce it into host cells (transformation/electroporation); (6) select transformed cells, grow them, and harvest the expressed product.
Answer: Restriction enzymes (restriction endonucleases) are bacterial enzymes that cut DNA at specific short recognition sequences (usually 4–8 base pairs, often palindromic). They evolved as bacterial defence against viruses. Cuts may be blunt or staggered, leaving sticky ends — single-stranded overhangs that can pair with matching ends. In cloning, the same enzyme cuts both gene and vector so their ends match for ligation. They also generate RFLPs: because individuals differ in restriction sites, their DNA gives different fragment patterns, used in DNA fingerprinting, paternity testing and disease diagnosis. Famous examples: EcoRI (from E. coli), HindIII.
Answer: PCR is an in-vitro method to amplify a specific DNA segment. Requirements: template DNA, two primers flanking the target, Taq DNA polymerase (heat-stable, from Thermus aquaticus), dNTPs, buffer and a thermal cycler. Cycle: denaturation (~94 °C, strands separate), annealing (~50–60 °C, primers bind), extension (72 °C, Taq builds new strands). Each cycle doubles the target; 25–35 cycles yield millions to billions of copies. Applications: medical diagnosis (infections, genetic disorders), forensic DNA fingerprinting, paternity testing, cloning, ancient-DNA studies and research. Its power is also its weakness: tiny contamination can be amplified, so strict sterile technique is essential.
Answer: Transformation: bacteria are made competent (calcium chloride + heat shock) so they take up plasmid DNA carrying the human gene. Electroporation: a brief high-voltage pulse opens temporary pores in the bacterial membrane, letting plasmid DNA enter. Conjugation/transduction can also transfer plasmids in research settings. After introduction, bacteria are grown on selective medium (antibiotic resistance on the plasmid) so only transformed cells survive; these are cultured in fermenters to produce the human protein (e.g. insulin).
Answer: Local-research task: students should identify genetic-engineering work in their own province/city — e.g. in Punjab, Pakistan: work on Bt cotton at agricultural research institutes, insulin and vaccine production in pharmaceutical units, and university biotechnology departments (e.g. Punjab University, NIBGE Faisalabad for crop biotechnology). Students should name the institution, the product or crop, and its benefit, from current official sources.
Answer: Method task: in DNA fingerprinting, every band in the child's profile must come from one of the two parents. If a tested man lacks several bands that the child has (and that the mother also lacks), he is excluded as the father; if all the child's non-maternal bands match him, paternity is supported with very high probability (the more markers tested, the stronger the conclusion). Courts use this with strict chain-of-custody and statistical reporting.
Answer: PCR applications: diagnosing infections (TB, hepatitis, COVID-type viruses) from tiny samples; detecting genetic mutations before symptoms; forensic identification from a hair or blood drop; paternity testing; checking food for contamination; cloning genes for research; and studying extinct organisms' DNA. Any job needing many copies of a specific DNA piece from almost nothing uses PCR.
Answer: The Human Genome Project (1990–2003) read all ~3 billion base pairs of human DNA — the complete instruction book for a human — using thousands of scientists in many countries and new sequencing technology. It was ambitious in scale (the largest biology project ever), cost, computing and cooperation, and it transformed medicine: it enabled gene-based diagnosis, personalised medicine, ancestry testing and the discovery of thousands of disease genes. Its data is freely shared and still drives research today.
