INTRODUCTION

Chromatography separates mixtures by how their components divide between two phases. This unit covers paper chromatography (Rf values), thin-layer chromatography (TLC), and gas chromatography (GC) — their principles, apparatus, measurements, uses in forensics, pharmaceuticals and food safety, and their limitations.

1. Principle: Two Phases and Partition
🏷 Tags: chromatography, stationary phase, mobile phase, partition, affinity
ENGLISH

Chromatography separates the components of a mixture because they distribute differently between a stationary phase (fixed) and a mobile phase (moving). Components that spend more time in the mobile phase travel further; components that stick to the stationary phase (by adsorption — sticking only to the surface of another material) travel less far.

Adsorption means a substance sticks only to the surface of another material (it does not dissolve into it or react with it). Example: a dye adsorbed on silica gel, or impurities adsorbed on charcoal. In paper chromatography the stationary phase is the water trapped in the paper fibres; the mobile phase is the solvent rising by capillary action.

Why must the solvent level be below the spot? So the sample does not dissolve directly into the solvent reservoir — it must travel up with the solvent front, separating as it goes. Why use pencil, not ink, for the baseline? Ink contains dissolved coloured components that would move with the solvent and interfere; pencil (graphite) is insoluble and stays put.

Chromatography principleStationary phase: fixed (water in paper, silica on TLC plate)Mobile phase: moving (solvent, carrier gas)More time in mobile phase → travels furtherAdsorption = sticking to a surface only
2. Paper Chromatography and Rf Valuesپیپر کرومیٹوگرافی اور Rf ویلیوز
🏷 Tags: paper chromatography, Rf value, solvent front, spot
ENGLISH

In paper chromatography, a spot of mixture is placed on a pencil baseline, the paper is dipped in solvent (solvent level below the spot), and the solvent rises, carrying components at different speeds. The result is a chromatogram — a pattern of spots.

Rf value = distance travelled by the component ÷ distance travelled by the solvent front. Rf has no units and is always between 0 and 1. A component more soluble in the mobile phase travels further (higher Rf); a component more soluble in the stationary phase (water in the paper) travels less far. A very low Rf means the substance sticks strongly to the water inside the paper (strong adsorption to the stationary phase), not that it is insoluble or dense.

What does a chromatogram provide? The number of components (number of spots), their identities (by comparing Rf values with known substances in the same solvent), and rough proportions (spot size/intensity).

Rf valueRf = distance of spot ÷ distance of solvent frontNo units, always between 0 and 1High Rf: more soluble in mobile phaseLow Rf: sticks strongly to stationary phase
🏷 Tags: TLC, silica, polarity, plate, separation
ENGLISH

In TLC, the stationary phase is a thin layer of solid (usually silica gel or alumina) coated on a glass or plastic plate. The plate is spotted on a baseline and placed in a solvent tank, exactly like paper chromatography — but TLC is considered faster than paper chromatography because it has a solid stationary phase that allows quicker separation.

Two advantages of TLC over paper chromatography: (1) faster separation; (2) sharper spots and a wider choice of stationary phases and solvents, so it separates substances paper cannot, and spots can be visualised under UV light. Polar substances move slowly in TLC because they adsorb strongly onto the polar silica stationary phase; a non-polar compound travels further up the plate because it is weakly adsorbed.

Why is TLC not suitable for large-scale separation? The plate is small and the layer is thin, so it can only handle tiny samples — it is an analytical technique, not a preparative one. With a more polar solvent, Rf values increase: components spend more time in the mobile phase and are less adsorbed.

TLC vs paperStationary phase: solid silica/alumina layerFaster than paper + sharper spotsPolar substances: move slowly (strong adsorption)Non-polar substances: travel further up the plate
4. Gas Chromatography (GC)گیس کرومیٹوگرافی (GC)
🏷 Tags: gas chromatography, carrier gas, retention time, chromatogram
ENGLISH

In gas chromatography, the mobile phase is an inert carrier gas (helium or nitrogen) and the stationary phase is a high-boiling liquid coated on a solid support inside a long, coiled column in an oven. The role of the carrier gas is to carry the vaporised sample through the column — it does not react with the sample and does not act as the stationary phase.

Components: injector port (sample vaporised), column (separation), oven (temperature control), detector (records the chromatogram). The stationary phase must have a high boiling point to remain liquid during the process — if it evaporated, there would be no stationary phase left. Samples must be volatile in GC because only gases/vapours can travel through the column; non-volatile samples cannot be analysed (a limitation), and very high temperatures can decompose samples (another limitation).

Measurements: retention time = the time from injection to the peak maximum — it identifies a substance (compared with standards). Peak area (or height, when base widths are similar) indicates the amount (proportion) of the substance — so if two peaks have similar base widths but different heights, composition is estimated by peak height.

Gas chromatographyMobile phase: inert carrier gas (He or N2)Stationary phase: high-boiling liquid on a supportRetention time → identity; peak area → amountSamples must be volatile (limitation)

ENGLISH

Applications: forensic science (ink, drugs, poisons), pharmaceuticals (purity, drug analysis), food safety (additives, contaminants), pollution detection, and amino-acid analysis. A chromatogram gives the number of components, their identities (Rf or retention time vs standards) and their proportions (spot intensity or peak area).

Comparison: paper — cheapest, slowest, water stationary phase. TLC — faster, sharper, solid stationary phase, small samples only. GC — fastest for volatile mixtures, quantitative (peak areas), but only for volatile, thermally stable samples and needs expensive apparatus.

Worked TLC example (exercise): an amino-acid mixture gives spots A and B. Spot A has Rf ≈ 0.71 → leucine (0.71); spot B has Rf ≈ 0.40 → alanine (0.40), using the table (alanine 0.40, glutamic acid 0.29, leucine 0.71, valine 0.61). With a more polar solvent, Rf values increase because components dissolve better in the mobile phase and adsorb less.

Three techniques comparedPaper: cheap, slow, water stationary phaseTLC: fast, sharp, small samples onlyGC: volatile samples, retention time + peak areaAll three: identify by comparison with standards

Section 2: Short Answer Questions
Q1. What is the principle of paper chromatography?

Answer: Components of a mixture distribute differently between the stationary phase (water trapped in the paper) and the mobile phase (solvent rising by capillary action). Components more soluble in the mobile phase travel further; those adsorbed more strongly travel less far.

Q2. Why is pencil used instead of ink in chromatography to mark the baseline?

Answer: Ink contains soluble coloured dyes that would dissolve and move with the solvent, interfering with the chromatogram. Pencil graphite is insoluble and stays on the baseline.

Q3. State two advantages of TLC over paper chromatography.

Answer: (1) Faster separation. (2) Sharper spots and a wider range of stationary phases/solvents, separating substances paper cannot; spots can be seen under UV light.

Q4. Why do polar substances move slowly in TLC?

Answer: They adsorb strongly onto the polar silica stationary phase, spending little time in the mobile phase, so they travel only a short distance.

Q5. Why is TLC not suitable for large-scale separation?

Answer: The plate and layer are small and thin, so only tiny samples can be applied — TLC is an analytical method, not a preparative one.

Q6. What is the function of injector port in GC?

Answer: To vaporise the liquid sample instantly (flash vaporisation) and introduce it into the carrier-gas stream entering the column.

Q7. Why must samples be volatile in GC?

Answer: Only vapours can be carried by the carrier gas through the column; non-volatile substances would stay in the injector and never separate.

Q8. State two limitations of gas chromatography.

Answer: (1) Only volatile, thermally stable samples can be analysed. (2) The apparatus is expensive, and compounds that decompose at high temperature cannot be separated.

Q9. What information does a chromatogram provide?

Answer: The number of components (number of spots/peaks), their identities (Rf values or retention times compared with standards), and their relative amounts (spot intensity or peak area).

Q10. What is adsorption? Give one example.

Answer: Adsorption is the sticking of a substance only to the surface of another material. Example: dye molecules adsorbed on silica gel in TLC, or impurities adsorbed on activated charcoal.

Section 3: Constructed Response Questions
Q1. A mixture of amino acids is analysed using thin layer chromatography. The chromatogram obtained is shown in the book. The table shows Rf values: alanine 0.40, glutamic acid 0.29, leucine 0.71, valine 0.61. i) Use the chromatogram and the Rf values to deduce the amino acid responsible for spot A and spot B. ii) A second chromatogram of the same mixture is taken using a more polar solvent. Predict the effect on the Rf values of the amino acids. Explain your reasoning.

Answer: i) Spot A (higher up, Rf ≈ 0.71) = leucine; spot B (lower, Rf ≈ 0.40) = alanine. ii) Rf values increase: a more polar solvent dissolves the amino acids better in the mobile phase and competes with the polar silica for them, so they adsorb less and travel further.

Q2. Gas chromatography involves a stationary phase and a mobile phase. a) Name a suitable substance that could be used for each phase. b) A mixture of three organic compounds is separated by gas chromatography. i) Explain the meaning of the term retention time. ii) Calculate the % of N in the mixture. Show your working.

Answer: a) Stationary phase: a high-boiling liquid such as silicone oil (or polyethylene glycol) coated on an inert support. Mobile phase: an inert carrier gas such as helium or nitrogen. b) i) Retention time is the time from injection until the peak maximum appears — it identifies a substance when compared with standards. ii) Using the printed peak areas (14, 25, 52, 38): total = 129; the 25-unit peak = 25/129 × 100 ≈ 19.4%. NOTE: one peak in the printed chromatogram has no labelled area, so this calculation uses only the four labelled areas.

Q3. An unknown amino acid is analysed by thin-layer chromatography. Two chromatograms of the unknown amino acid and four reference amino acids, P, Q, R and S, are obtained using two different solvents. Identify the unknown amino acid. Justify your answer.

Answer: The unknown is the reference amino acid whose spot height matches the unknown in BOTH solvents — a single solvent can give a coincidence, but matching in two different solvents confirms identity. From the printed figures the unknown matches reference R (the only reference coinciding with the unknown in the first solvent and closest in the second); the printed spot positions are small, so the method — matching in both solvents — is the reliable identification, not a single-solvent guess.

Section 4: Descriptive (Long) Questions
Q1. Describe the principle and working of paper chromatography.

Answer: Principle: partition between the stationary phase (water in paper fibres) and the mobile phase (solvent). Q2. Explain the principle and working process of thin layer chromatography (TLC) in detail.

Answer: Principle: adsorption on a solid stationary phase (silica/alumina layer) vs solubility in the mobile phase. Working: coat a plate, spot the sample on a baseline, develop in a solvent tank, dry, visualise spots (UV light or iodine), measure Rf values and compare with standards. Advantages over paper: faster, sharper spots, wider solvent choice. Limitation: analytical scale only.

Q3. Describe the main components of gas chromatography.

Answer: Carrier-gas supply (He/N₂, inert); injector port (flash vaporisation); column (long coiled tube with high-boiling liquid stationary phase on a support, in an oven); oven (precise temperature control); detector (e.g. flame ionisation detector) producing the chromatogram; recorder/computer. Retention time identifies, peak area quantifies.
