1. Principle: Nuclear Spin and TMSاصول: نیوکلیئر اسپن اور TMS
🏷 Tags: NMR, nuclear spin, TMS, standard, resonance, magnetic field
ENGLISH

NMR relies on the magnetic properties of nuclei known as nuclear spin. Nuclei such as ¹H and ¹³C behave like tiny magnets. In a strong magnetic field they align with or against the field, and radio-frequency radiation flips them between these states. The exact frequency needed depends on the nucleus's chemical environment — that is the basis of NMR.

TMS (tetramethylsilane, Si(CH₃)₄) is the standard for ¹H NMR for two reasons: (1) its 12 protons are all equivalent, giving a single sharp peak; (2) it is chemically inert and volatile (easily removed), and its peak appears at the extreme upfield end (defined as δ = 0 ppm) without overlapping other signals. Chemical shift (δ) is measured relative to TMS and expressed in parts per million (ppm) — not in tesla, hertz or degrees Celsius.

NMR basicsNMR relies on nuclear spin (1H, 13C)TMS standard: 12 equivalent protons, single peak at δ = 0TMS: inert, volatile, peak at extreme upfield endChemical shift δ measured in ppm (relative to TMS)
2. Chemical Shift and Peak Areas
🏷 Tags: chemical shift, peak area, integration, environment, δ
ENGLISH

Chemical shift (δ) tells us about a proton's chemical environment: protons near electronegative atoms or π systems are deshielded and appear at higher δ (downfield, left); protons in electron-rich environments are shielded and appear at lower δ (upfield, right). Typical ranges: alkyl C–H ~0.9–1.7 ppm, C–H next to O/N/halogen ~3–4 ppm, aldehyde H ~9–10 ppm, carboxylic-acid O–H ~11 ppm.

The primary purpose of measuring peak area in ¹H NMR is to find the relative number of protons in each environment. Example: a compound C₅H₁₀O with a peak-area ratio of 1:3:6 has 1 + 3 + 6 = 10 protons in total, so the peaks represent 1H, 3H and 6H respectively. In a low-resolution spectrum you use peak count and area ratio; in a high-resolution spectrum you also use splitting.

3. Splitting: The n+1 Ruleاسپلٹنگ: n+1 رول
🏷 Tags: splitting, n+1 rule, neighbour, doublet, triplet, quartet
ENGLISH

Splitting happens because neighbouring protons' spins slightly change the magnetic field felt by a proton. The n+1 rule: a signal splits into (n+1) peaks, where n is the number of protons on the adjacent carbon atoms (not the whole molecule, not the carbon being analysed, not the signal's area).

Examples: a proton with 2 neighbouring protons (n=2) gives a triplet. The –CH₂– signal in ethanol (next to –CH₃, 3 protons) splits into a quartet. A –CH₂– group lying between a –CH₃ and a –CH< group has 3 + 1 = 4 neighbours → a quintet (5 peaks). Splitting is typically not observed for O–H or N–H protons because of rapid chemical exchange: the proton swaps between molecules so fast that the coupling is averaged out, and the peak appears as a singlet.

4. Deuterated Solvents and the D₂O Shakeڈیوٹریٹڈ سالونٹس اور D₂O شیک
🏷 Tags: deuterated solvent, CDCl₃, D₂O shake, exchangeable proton
ENGLISH

Deuterated solvents (e.g. CDCl₃) are used in ¹H NMR to avoid solvent signal interference — ordinary solvents contain protons that would swamp the spectrum. Deuterium (D) does not appear in a ¹H NMR spectrum because it is a different nucleus with a different resonance frequency; the spectrometer is tuned to ¹H only.

D₂O shake test: add a few drops of D₂O to the sample and re-run the spectrum. Exchangeable protons (O–H, N–H) swap with deuterium: R–OH + D₂O ⇌ R–OD + HOD. Because deuterium resonates at a different frequency, the O–H/N–H peak disappears from the ¹H NMR spectrum. This is how you confirm a sample contains an O–H group: the peak that vanishes after the shake is the exchangeable proton.

🏷 Tags: carbon-13 NMR, peak count, structure deduction, symmetry
ENGLISH

In ¹³C NMR, the number of peaks equals the number of chemically non-equivalent carbons (not the total carbon atoms, not the hydrogens). The chemical shift of a ¹³C peak identifies the functional-group environment (C=C, C=O, C–O, etc.). ¹³C spectra are usually proton-decoupled, so each carbon gives a singlet.

Examples: propane (CH₃CH₂CH₃) shows 2 peaks (the two equivalent CH₃ carbons, and the central CH₂). Pentane shows 3 peaks (two equivalent terminal CH₃, two equivalent CH₂, one central CH₂). 2,2-dimethylpropane shows 2 peaks (four equivalent CH₃ carbons, and the central quaternary carbon).

Structure deduction from ¹H NMR: butanoic acid (CH₃CH₂CH₂COOH) shows: CH₃ triplet (3H, ~0.9 ppm), CH₂ sextet (2H, ~1.6 ppm, 5 neighbours), CH₂ triplet (2H, ~2.3 ppm, next to C=O), and COOH singlet (1H, ~11 ppm, disappears on D₂O shake). A spectrum with only a 3H triplet (~1.2 ppm) and a 2H quartet (~3.7 ppm) fits chloroethane, CH₃CH₂Cl. A compound C₄H₈O that gives a positive 2,4-DNPH test (carbonyl), a positive iodoform test (CH₃CO–) and a negative Fehling's test (not an aldehyde) is butan-2-one (CH₃COCH₂CH₃: 3H singlet, 2H quartet, 3H triplet).

Section 2: Short Answer Questions
Q1. State two reasons why TMS is a standard for ¹H NMR.

Answer: (1) All 12 protons are equivalent, giving a single sharp peak. (2) It is chemically inert and volatile, and its peak (δ = 0) sits at the upfield end without overlapping other signals.

Q2. Why does deuterium (D) not appear in a ¹H NMR spectrum?

Answer: Deuterium is a different nucleus (²H) with a different resonance frequency; a ¹H NMR spectrometer is tuned to ¹H only, so D is invisible in it.

Q3. A compound (C₅H₁₀O) has a ¹H NMR peak area ratio of 1:3:6. How many protons does each signal represent?

Answer: Total parts = 1 + 3 + 6 = 10, and the formula has 10 protons, so the signals represent 1H, 3H and 6H respectively.

Q4. What is the purpose of the D₂O shake test in ¹H NMR?

Answer: To identify exchangeable protons: after adding D₂O, O–H and N–H protons exchange for deuterium and their peaks disappear from the ¹H NMR spectrum, confirming an –OH or –NH group.

Q5. List the key features used to interpret a ¹H NMR spectrum.

Answer: Number of peaks (environments), chemical shift δ (environment type), peak area/integration (relative proton numbers), and splitting pattern with the n+1 rule (neighbouring protons); plus the D₂O shake for exchangeable protons.

Q6. What does the number of peaks in a ¹³C NMR spectrum represent?

Answer: The number of chemically non-equivalent carbon environments in the molecule.

Section 3: Constructed Response Questions
Q1. A -CH₂- group is lying between a -CH₃ and a -CH< group. What is the splitting pattern for the -CH₂- signal? Show your calculation.

Answer: Neighbours = 3 (from CH₃) + 1 (from CH) = 4. Splitting = n+1 = 5 → a quintet (five peaks, intensity ratio 1:4:6:4:1).

Q2. Predict the ¹H NMR spectrum of 1-butanoic acid.

Answer: Butanoic acid CH₃CH₂CH₂COOH: CH₃ triplet (3H, ~0.9 ppm); CH₂ sextet (2H, ~1.6 ppm, 5 neighbours: CH₃ + CH₂); CH₂ triplet (2H, ~2.3 ppm, next to C=O); COOH singlet (1H, ~11 ppm, disappears on D₂O shake).

Q3. An organic compound has a molecular formula C₂H₅Cl. The ¹H NMR of the compound is given in the book. Deduce the structure of the compound by using its spectrum. Explain your working in detail.

Answer: NOTE: the book prints the formula in garbled form ("C₃H₂Cl"); the spectrum shown — a 3H triplet near 1.2 ppm and a 2H quartet near 3.7 ppm, totalling 5H — fits C₂H₅Cl. Working: 3H triplet means a CH₃ next to a CH₂; 2H quartet means a CH₂ next to a CH₃, deshielded to ~3.7 ppm by chlorine. Structure: CH₃CH₂Cl (chloroethane).

Q4. An organic compound has a formula C₄H₈O. In laboratory, the compound gives 2,4-DNPH and iodoform tests positive but Fehling's test shows a negative result. The ¹H NMR of the compound is shown in the book. Deduce the structure of the compound and justify your answer.

Answer: 2,4-DNPH positive → carbonyl present. Iodoform positive → CH₃CO– group present. Fehling's negative → not an aldehyde, so it is a ketone: butan-2-one, CH₃COCH₂CH₃. NMR confirms: CH₃CO singlet (3H, ~2.1 ppm), CH₂ quartet (2H, ~2.7 ppm), CH₃ triplet (3H, ~1.0 ppm).

Section 4: Descriptive (Long) Questions
Q1. A chemist suspects a sample contains an O-H group. a) Describe the "D₂O shake" procedure. b) What is the expected observation in the ¹H spectrum after the shake? c) Write an equilibrium equation for the exchange and explain why it causes this observation.

Answer: a) Add a few drops of D₂O to the NMR sample, shake, and re-record the spectrum. b) The O–H peak disappears (or greatly shrinks). c) R–OH + D₂O ⇌ R–OD + HOD. The proton is replaced by deuterium, which resonates at a different frequency and is invisible in ¹H NMR, so the signal vanishes.

Q2. Predict the number of ¹³C NMR peaks for the following two isomers: a) Pentane b) 2,2-dimethylpropane. Justify each answer by identifying the sets of equivalent carbons.

Answer: a) Pentane: 3 peaks — two equivalent terminal CH₃ carbons (one peak), two equivalent CH₂ carbons (one peak), and the central CH₂ (one peak). b) 2,2-dimethylpropane: 2 peaks — four equivalent CH₃ carbons (one peak) and the central quaternary carbon (one peak).
