1. Drug Safety: Therapeutic Index and Window
🏷 Tags: therapeutic index, therapeutic window, ED₅₀, TD₅₀, drug safety
ENGLISH

Therapeutic Index (TI) = TD₅₀ ÷ ED₅₀. TD₅₀ is the dose causing a toxic effect in 50% of people (not the lowest harmful dose, not the dose effective for 100%). ED₅₀ is the dose effective in 50% of people. A high TI means a wide safety margin — a safer drug.

Therapeutic Window (TW) is the range between the minimum effective concentration (MEC) and the minimum toxic concentration (MTC): TW = MTC − MEC. A wider window is safer. Worked comparison: Drug A: ED₅₀ = 20 mg, TD₅₀ = 80 mg → TI = 80 ÷ 20 = 4. Drug B: MEC 5 mg/L, MTC 50 mg/L → window 45. Drug C: effective 10 mg, toxic 15 mg → window 5. So Drug B has the wider therapeutic window (45 vs 5), and Drug A is relatively safer than Drug C (TI of 4 vs a toxic/effective ratio of only 1.5).

Drug safety numbersTI = TD50 ÷ ED50 (higher = safer)Therapeutic window = MTC − MEC (wider = safer)Drug A: TI = 80 ÷ 20 = 4Drug B window 45 vs Drug C window 5 → B wider
2. How Drugs Work: Penicillin and Opiates
🏷 Tags: penicillin, opiate, mechanism, selective target, morphine
ENGLISH

Penicillin: its key structural feature is the four-membered beta-lactam ring. The strained ring binds and inactivates transpeptidase, the enzyme that cross-links bacterial cell walls — so the wall cannot be completed and the bacterium dies. Humans are safe because we have no cell walls and no transpeptidase target.

Opiates like morphine reduce pain because they bind to opioid receptors, blocking pain signals — they do not destroy pain signals or build new brain cells; they mimic the body's own endorphins at specific receptors. pH in the stomach matters: many drugs are weak acids or bases, so stomach pH decides how much is ionised and absorbed — a drug that is ionised is poorly absorbed through membranes.

Why are safe, effective antivirals harder to develop than antibiotics? Viruses replicate inside human cells using the host's own machinery, so a drug that attacks the virus usually harms the host cell too; bacteria, by contrast, have their own distinct structures (cell walls, ribosomes) that drugs can target selectively.

Drug mechanismsPenicillin: beta-lactam ring → blocks transpeptidaseMorphine: binds opioid receptors → blocks pain signalsAntibiotics target bacterial-only structuresAntivirals are hard: viruses use host machinery
3. Fertilisers, Pesticides and Soil
🏷 Tags: fertiliser, NPK, pesticide, resistance, soil salinization
ENGLISH

NPK — the three primary macronutrients: Nitrogen (N) promotes protein synthesis and chlorophyll (green growth); Phosphorus (P) is needed for energy transfer (ATP), root growth and flowering; Potassium (K) activates enzymes, controls water and improves disease resistance. Synthetic fertilisers are manufactured chemicals of known composition (fast-acting, precise); organic fertilisers come from plant/animal matter (slow-release, improve soil structure).

Pesticide resistance: repeated application of the same pesticide selects for resistant pests — susceptible individuals die, resistant ones survive and breed (natural selection), so the population becomes resistant and the pesticide stops working. Rotating pesticides and integrated pest management slow this down.

Soil salinization: in hot regions with low rainfall, irrigation water evaporates and leaves dissolved salts behind in the topsoil; the salt builds up, damages roots by osmosis and makes the soil infertile. Acid rain damages soil mainly by leaching essential nutrients and releasing toxic Al³⁺ (it does not raise soil pH — it lowers it).

NPK functionsN: proteins + chlorophyll (green growth)P: ATP energy, roots, floweringK: enzymes, water control, disease resistanceResistance: natural selection under repeated spraying
4. Climate Change, Acid Rain and Crops
🏷 Tags: climate change, acid rain, crop, greenhouse, pollution
ENGLISH

A primary effect of climate change on agriculture is reduced yields due to heat stress and erratic rainfall — not increased yields globally, not reduced CO₂, and not fewer extreme weather events. Heat damages flowering and grain-filling; erratic rain causes drought and flood in turn.

Acid rain forms when SO₂ and NOₓ from burning fossil fuels dissolve in rainwater (→ sulphuric and nitric acids). Effects: leaches nutrients (Ca²⁺, Mg²⁺, K⁺) from soil, releases toxic Al³⁺ that damages roots, damages leaves and buildings, and acidifies lakes. Genetic engineering benefits: pest-resistant, drought-tolerant and higher-yield crops; risks: gene flow to wild relatives, loss of biodiversity, pest resistance, and unknown long-term effects.

Acid rain pathwaySO2 + NOx from fuel burning → acids in rainLeaches nutrients, releases toxic Al3+Damages roots, leaves, buildings, lakesClimate change: heat stress + erratic rain → lower yields
5. Pakistan: Raw Materials, Industry and Safety
🏷 Tags: Pakistan, raw material, halite, natural gas, industry, safety
ENGLISH

Pakistan's key raw materials: rock salt (halite) from Khewra and other mines, limestone (cement), gypsum, coal, natural gas, and cotton. In Pakistan, rock salt (halite) is a key raw material for producing caustic soda and chlorine (by the chlor-alkali process, electrolysis of brine: 2NaCl + 2H₂O → 2NaOH + Cl₂ + H₂). Ammonia gas in industry is prepared by the Haber–Bosch process (N₂ + 3H₂ → 2NH₃). Plastics, synthetic fibres and synthetic rubber are primary products of the petrochemical industry.

Urea from natural gas: CH₄ + air → (steam reforming) hydrogen → (Haber–Bosch) ammonia — ammonia is the crucial intermediate synthesised from CH₄ and air — then NH₃ + CO₂ → urea. Other sectors: textiles rely on dyes and finishing chemicals; construction relies on cement and steel; cosmetics/perfumes use esters (from a carboxylic acid + an alcohol, acid catalyst).

Safety: the primary purpose of ventilation in a chemical facility is to remove toxic fumes. A corrosive hazard destroys living tissue and materials on contact (e.g. concentrated sulphuric acid); a toxic hazard poisons by ingestion, inhalation or skin absorption (e.g. cyanides, methanol). Highly oxidising chemicals (concentrated HNO₃) must be stored separately from flammable organics (ethanol) because together they can ignite. Safety checklist: PPE, ventilation, labelling, separate storage of incompatibles, eyewash/safety shower, spill kits, emergency exits, training.

Section 2: Short Answer Questions
Q1. Write significance of Therapeutic Window and how it differs from Therapeutic Index.

Answer: The therapeutic window (MTC − MEC) is the practical safe dosing range for a patient. The therapeutic index (TD₅₀ ÷ ED₅₀) is a population ratio of toxic to effective doses. TI compares two population doses; TW gives the concentration range within which a drug is both effective and safe.

Q2. How structure of penicillin allows it to inhibit the transpeptidase enzyme?

Answer: Penicillin's strained four-membered beta-lactam ring mimics the D-Ala-D-Ala end of the bacterial cell-wall precursor, so it fits the transpeptidase active site and binds it irreversibly, blocking the cross-linking of the wall.

Q3. What is the primary mechanism of action for opiate drugs, such as morphine, in providing analgesia (pain relief)?

Answer: Opiates bind to opioid receptors (mainly μ receptors) in the brain and spinal cord, mimicking endorphins, and block the transmission of pain signals.

Q4. Why is it significantly more challenging to develop safe and effective antiviral drugs compared to antibacterial drugs (antibiotics)?

Answer: Viruses replicate inside host cells using host machinery, so drugs that stop the virus tend to harm the host cell too. Bacteria have unique targets (cell walls, bacterial ribosomes) absent in human cells, allowing selective attack.

Q5. What is the concept of pesticide resistance in a target pest population, linking it to the principles of natural selection and repeated application?

Answer: Repeated spraying kills susceptible pests but spares rare resistant mutants; the survivors breed, so each generation has more resistant individuals until the population is largely resistant and the pesticide fails. This is natural selection in action.

Q6. Define soil salinization. How it can be caused by irrigation practices in regions experiencing higher temperatures and reduced precipitation.

Answer: Soil salinization is the build-up of soluble salts in topsoil. In hot, dry regions, irrigation water evaporates quickly, leaving its dissolved salts behind; with little rain to wash them away, salts accumulate, damaging roots and reducing fertility.

Q7. How does the chemical industry contribute to both energy production from fossil fuels and the development of renewable energy technologies?

Answer: From fossil fuels: refining, cracking and petrochemicals produce fuels, plastics and solvents. For renewables: the industry makes silicon for solar panels, materials for wind-turbine blades, batteries for storage, and catalysts for green hydrogen.

Q8. Name the industrial process used to produce caustic soda (NaOH) and chlorine (Cl₂) from the rock salt (NaCl).

Answer: The chlor-alkali process: electrolysis of concentrated brine, 2NaCl + 2H₂O → 2NaOH + Cl₂ + H₂.

Q9. State two different types of products (other than fuels) that are derived from petrochemical industry, linking them to their use in daily life.

Answer: Plastics (packaging, bottles, pipes) and synthetic fibres (polyester clothing, carpets). Also: synthetic rubber (tyres), detergents, paints and solvents.

Q10. Distinguish between a corrosive hazard and a toxic hazard, giving one example of a chemical for each.

Answer: Corrosive: destroys tissue/materials on contact (e.g. concentrated sulphuric acid burns skin). Toxic: poisons the body after ingestion, inhalation or absorption (e.g. cyanide, methanol) without necessarily corroding tissue.

Section 3: Constructed Response Questions
Q1. Drug A has an ED₅₀ of 20 mg and a TD₅₀ of 80 mg. Drug B has a Minimum Effective Concentration (MEC) of 5 mg/L and a Minimum Toxic Concentration (MTC) of 50 mg/dm³. Drug C has a therapeutic window where the effective dose is 10 mg and the toxic dose is 15 mg. (i) Define Therapeutic Index (TI) and calculate the TI for Drug A. (ii) Define Therapeutic Window (TW) and identify which drug (B or C) has the wider therapeutic window. Justify your answer. (iii) Compare the relative safety of Drug A and Drug C.

Answer: i) TI = TD₅₀ ÷ ED₅₀ = 80 ÷ 20 = 4. ii) TW = MTC − MEC. Drug B: 50 − 5 = 45. Drug C: 15 − 10 = 5. Drug B has the wider window (45 vs 5), so it is safer to dose. iii) Drug A (TI = 4) is relatively safer than Drug C (toxic/effective ratio 15 ÷ 10 = 1.5) — Drug C's toxic dose is dangerously close to its effective dose. جواب: i) TI = 80 ÷ 20 = 4۔ ii) دوا B کی ونڈو 45، دوا C کی 5 — B زیادہ چوڑی۔ iii) دوا A (TI = 4) دوا C (1.5) سے محفوظ ہے۔

Q2. (i) Contrast synthetic and organic fertilizers based on their origin. (ii) Identify the three primary macronutrients (N, P, K) and describe the specific biological function of each in a plant.

Answer: i) Synthetic fertilisers are manufactured chemicals (e.g. ammonium nitrate, urea) of known, concentrated composition, fast-acting. Organic fertilisers come from plant/animal matter (manure, compost), release nutrients slowly and improve soil structure. ii) N: proteins and chlorophyll (leafy growth). P: ATP energy transfer, roots, flowering and seeds. K: enzyme activation, water regulation, disease resistance.

Q3. Describe the chemical pathway from Natural Gas (CH₄) to urea fertilizer. Your answer must identify the crucial intermediate compound that is synthesized from CH₄ and air.

Answer: CH₄ + steam → (steam reforming) H₂ + CO. H₂ + N₂ (from air) → (Haber–Bosch, Fe catalyst) NH₃ — ammonia is the crucial intermediate synthesised from CH₄ and air. NH₃ + CO₂ → (high pressure) urea, CO(NH₂)₂. So the pathway is: natural gas → hydrogen → ammonia → urea.

Section 4: Descriptive (Long) Questions
Q1. Describe the structure and mechanism of action of penicillin.

Answer: Structure: a beta-lactam ring (four-membered) fused to a thiazolidine ring, with a variable side chain. Mechanism: the strained beta-lactam ring mimics the D-Ala-D-Ala substrate of transpeptidase, binds the enzyme's active site irreversibly, and blocks cross-linking of peptidoglycan in the bacterial cell wall — the wall weakens, the cell swells and bursts. Human cells lack walls, so they are unaffected.

Q2. What is Genetic Engineering? Give its main benefits and risks.

Answer: Genetic engineering is the direct manipulation of an organism's DNA (e.g. inserting a gene for pest resistance into a crop). Benefits: pest-resistant, drought-tolerant, higher-yield crops; medicines like insulin. Risks: gene flow to wild relatives, reduced biodiversity, development of resistant pests, unknown long-term ecological and health effects.

Q3. Explain the role of industrial chemistry in economy of Pakistan. Also account for the main raw materials available in Pakistan for these industries.

Answer: Industrial chemistry supplies fertilisers (food security), cement and steel (construction), textiles (exports), pharmaceuticals, and petrochemicals (plastics, fuels). Pakistan's raw materials: rock salt (chlor-alkali), limestone and gypsum (cement), coal and natural gas (energy, fertilisers via ammonia/urea), cotton (textiles), and sugarcane by-products. These industries provide jobs, exports and import substitution.

Q4. Write a note on various industrial chemical processes. Also account for the applications of these chemical industries in Pakistan.

Answer: Haber–Bosch (ammonia → urea fertilisers), Contact process (sulphuric acid for fertilisers, detergents), chlor-alkali (NaOH + Cl₂ for soap, paper, water treatment), Solvay (sodium carbonate for glass, detergents), fractional distillation and cracking (fuels, petrochemicals). In Pakistan these underpin agriculture, textiles, construction, water treatment and consumer goods.

Q5. What are important safety measures which must be followed while working in different chemical industries? Make a checklist to verify their adoption.
